No. Having sex will not cause real permanent changes in the labia or vagina. You may experience some changes throughout the lifespan that are natural. Some women may see changes in their labia (as noted in this post) or vagina. Having lots of sex will not increase those changes.

There may be temporary growth during arousal and during sex when the blood is flowing to the genitals. At this time things might appear bigger or fuller than normal as they swell up. When you’re done having sex, that swelling will go down, and your genitals will return to their normal.

Having more prominent labia minora can be stressful for women because often times pornography only portrays one particular kind of woman, with small and tucked labia. The reality is that the vulva is different on all women, and it is perfectly normal and common to have labia minora that are not totally hidden away.
